C-Reactive Protein (CRP):

Procedure:

-10 µ serum + 10 µ reagent of CRF (على الشريحة السوداء) then mix with
rotation by hands for 3 min.
If -ve so the result is -ve in report
If +ve the titre is 6 mg/dl and we will repeat by the following values:

-The serum diluted by saline by the ratio 1 : 1 [mix1] .
Then take 10 µ of mix with 10 µ of reagent and rotate for 3 min.
If -ve so the result 6 mg/dl .
If +ve so the titre 12 mg/dl and repeat by the following values :

-The mix will diluted by saline by the ratio 1 : 1 or the serum diluted
by saline by ratio
1 serum : 2 saline [mix2].
Then take 10 µ of mix2 with 10 µ reagent and rotate for 3 min.
If -ve so the result 12 mg/dl .
If +ve so the titre 24 mg/dl and repeat by the same steps.

-Titre of this test : 6 , 12 , 24 , 48
If the result leads to more than 48 the report is more than 48 and not
higher titre values .

IMPORTANT COMMENT:

When testing new born children false -ve result are possible .

Expirement 5 : C-reactive protien (CRP) Monday 26-4-2010

BIOREX
Introduction :

CRP is used as marker of inflammation . Normally there is no CRP in
blood strem .

An increasing amount of CRP in the blood suggest that a patient has an
acute infection or inflammation .

Principle of method :

The CRP latex test is a rapid slied agglutination test for the
qualitative and semi-quantitative detection of CRP in serum.

The reagent containing particles coated with specific anti-human CRP
antibodies , agglutinates in the presence of CRP in the patient's serum .

Procedure contents :

1- R1 : CRP latex

2- R2 : positive control

3- R3 : negative control

4- slied reaction

5- Normal saline for dilution

6- pipette 50 ul & tips

7- fresh clear serum is used

Procedure :

* Qualitative test

- Reagent and sample to room
temperature is brined before use

- A drop (50ul) of sample , positive control , negative control are placed
on the reaction slied

- A drop of latex reagent to each test field are added

- we rotated the slied for 2 minutes and read
immediately under direct light .

Result :

The sample was negative for qualitative test

" negative reaction is indicate by a uniform
milky suspension with no agglutination as observable while positive reaction is
indicate by any observable agglutination in the reaction mixture " .

* Quantitative test (just to know but didn't applied)

- by using a saline the sample
is diluted 1:2 , 1:4 , 1:8 , 1:16 as needed

- A drop (50ul) of sample , positive control , negative control are placed
on the reaction slied

- A drop of latex reagent to each test field are added

- we rotated the slied for 2 minutes and read
immediately under direct light .

Result :

-The titer of the serum is
equivalent of the highest dilution which give positive reaction

Interpretation
of results :

- negative
reaction indicate there aren't CRP in serum , while positive results may indicate an acute infection or
inflammation , result above 1mg/dl is usually considered high for CRP .
